چکیده

Thymic stromal lymphopoietin (TSLP) is an epithelial cell derived cytokine belonging to the IL-7 family and a key initiator of allergic inflammation. Two main isoforms TSLP, classified as long- (lfTSLP) short-form (sfTSLP), have been reported in human, but their expression patterns role(s) cancers are not yet clear. mRNA was examined by isoform-specific RT-PCR RNA situ hybridisation. Epigenetic regulation investigated chromatin immunoprecipitation-PCR bisulfite sequencing. Tumour progression gene overexpression, viability assay, cancer organoid culture transwell invasion. Signals were proteome profiler protein array RNA-sequencing. With use primers probes, we uncovered that only sfTSLP expressed lines tumour tissues human ovarian endometrial cancers. We also showed epigenetic sfTSLP: transcription regulated histone acetylation at promoters cells, whereas silencing transcripts promoter DNA methylation cells. In vitro study ectopically overexpressing promoted growth Human phosphokinase application demonstrated overexpression activated phosphorylation multiple intracellular kinases (including GSK3α/β, AMPKα1, p53, AKT1/2, ERK1/2 Src) cells context-dependent manner. further impact on transcriptome RNA-sequencing found EFNB2 PBX1 downregulated suggesting role sfTSLP-mediated growth. conclusion, predominantly promotes

Thymic stromal lymphopoietin and cancer.

Originally shown to promote the growth and activation of B cells, thymic stromal lymphopoietin (TSLP) is now known to have wide-ranging effects on both hematopoietic and nonhematopoietic cell lineages. These include dendritic cells, basophils, mast cells, B cells, epithelial cells, and CD4(+), CD8(+), and NK T cells. Although TSLP's role in the promotion of Th2 responses has been studied extens...

Thymic stromal lymphopoietin is produced by dendritic cells.

Thymic stromal lymphopoietin (TSLP) is a type 1 cytokine that contributes to lymphopoiesis and the development of asthma and atopic dermatitis. TSLP acts on multiple lineages, including dendritic cells (DCs), T cells, NKT cells, eosinophils, and mast cells, mediating proliferation and survival and linking innate and adaptive immune responses. TSLP is produced by a range of cells, including epit...
